These Grow Kits now feature a groundbreaking process that involves starting with sterile grainspawn. In this method, customers will utilize aseptic practices found within home mycology to inoculate the grainspawn using a 10mL syringe of liquid culture mycelium. Each grainspawn bag comes equipped with a self-healing injection port, ensuring a seamless and contamination-resistant process. Please follow the instructions below for a successful cultivation journey.
Before you begin, ensure you have the following items ready:
-Pair of Nitrile gloves
-Dust mask or equivalent
-70% Isopropyl alcohol or disinfectant spray
-Papertowel
Setting Up Your Lab Space
Select a draft-free room, such as a bedroom or kitchen, to serve as your cultivation space. Minimize air currents by shutting off fans and heating. Keep doors closed to lower the risk of contaminants entering the Microppose Monotub Grow Kit during exposure to open air environments.
Preparing Your Syringe
Clean the surface you're working on, including the outside of the grainspawn bag and the self-healing injection port. Once the disinfectant has dried, attach the needle to your syringe, ensuring a complete attachment before removing the syringe cap. Remove the syringe cap and use a lighter to heat the metal syringe tip to red hot. Stop heating once red, avoiding overheating to prevent damage to the plastic portion of the syringe.
Allow the syringe tip to cool from red hot. DO NOT WIPE OR TOUCH THE NEEDLE WITH ANYTHING. After cooling, wait about 3 seconds, then insert it into the rubber injection port.
Once inside the port, inject 5-10cc of genetic material into the bag. If using only 1 bag for your syringe, you can inject up to 10cc, though this is not totally necessary.
Withdraw your syringe, recap the needle, and if reusing your genetic material syringe, replace the syringe cap. If not reusing, dispose of your needle appropriately, and place the syringe into recycling.
Incubation and Colonization
Allow the grainspawn to initially incubate for approximately 7 days. Mycelium should become visible within this period. Allow an additional 7-10 days for further growth before shaking the bag to homogenize and improve mycelium contact points. Be mindful not to handle the filter patch, this can be a problem area for introducting contaminants. After 21 days, shake again and let it finish growing in an incubation temperature range of 21-24°C. If the timelines are longer, be patience, there are many factors that play into the development and consolidation of the spawn.
Cleaning and Lab Setup
Once the colonization period is complete, its time to wipe the Microppose Monotub Grow Kit with disinfectant and place the liner evenly across the bottom of the tub. Install 2" Microppose PLUGS over the holes on each end, and the sides of the tub. Now break up the mycelium inside the spawnbag, cut the top off; and pour the contents onto the liner. Homogenize both spawn and substrate by mixing with sanitized gloved hands or utensils.
Smooth down the surface like you are battering up a cake. secure the lid, and place the kit in in an undisturbed environment, away from direct sunlight.
Mycelium Run and Fruiting
Mycelium growth and colonization typically start within 48 hours. If not, be patient as growth may be under the surface or conditions may not be ideal. Consider your temperature rage at this point to ensure 70-76F is being met. Consolidation of the substrate takes around 10-12 days on average. After this timeframe, you can remove the PLUGS and install the Microppose Air Exchange Filters. This will initiate the mushroom pins to begin forming.
Allow 24 hours between opening or burping the Microppose Monotub Grow Kit again.
Check the kit at least once daily during the fruiting stage. Evaporation triggers mushroom pinning but maintain humidity for optimal results (90%RH inside the bin is ideal). Replace lost humidity with very light misting when needed. No need to mist if there is water beading and moisture on the surface.
After 72-96 hours, small knotting of the mycelium will occur, followed by primordial forming mycelium, which will develop into 'pins' or the beginning stage of your mushrooms.
Harvesting and Rehydration
When mushrooms are matured, harvest them before the caps fully open. Check for contamination before opening the kit. Use alcohol-sanitized scissors or a scalpel to cut mushrooms close to the base.
After harvesting, rehydrate the kit by pouring 2 cups of COLD WATER into the substrate or misting the surface heavily. Allow 4-6 hours of resting before draining off excess water, or misting further.
The Microppose Monotub from this kit can be reused consecutively with any variety of mushrooms in your mycology journey.
