Agar Plates for Ra Mushrooms Canada Cultivation

Agar Plates consist of a mixture of water, agar, and vital nutrients, forming a gel-like substance that's an optimal medium for fungi. Beyond fungi, this medium also fosters growth for molds, yeasts, and various other microorganisms. Rest assured, the Agar Plates you've procured have undergone sterilization in a top-tier commercial steam autoclave, ensuring a completely sterile environment devoid of living organisms or spores.

Crucial Notes on ContaminationAgar Plates, given their nutrient-rich composition, are inherently prone to contamination from airborne molds, bacteria, and yeasts. When initiating growth using spore syringes, spore prints, or liquid cultures, the risk of contamination looms. One potential strategy to circumvent this challenge is through multiple agar-to-agar transfers, aiming for a pure culture.

Sources of Contamination to Be Mindful Of:

  1. Inoculum: While sourcing from a reputable provider can minimize risks, contamination remains a possibility.
  2. Clone Specimens: Cloning from either cultivated or wild mushrooms may introduce contaminants, emphasizing the need for meticulous transfers on agar.
  3. Tools: Whether scalpels or inoculation loops, always sterilize these instruments. Flame sterilization or pressure cookers are recommended.
  4. Airborne Contaminants: A square meter of air can contain thousands of fungal spores. Utilizing a Still Air Box (SAB) or flow hood is paramount.
  5. Personal Contamination: Skin and hair harbor various microorganisms. Prioritize wearing sterilized nylon gloves and uphold impeccable hygiene standards.
  6. Sealing: Seal agar plates with Parafilm meticulously, ensuring no gaps or holes.
  7. Construction of a Still Air Box (SAB): A clear 56 qt plastic tote, fitted with hand-sized holes, can serve as an efficient SAB. Regularly sanitize its interior using isopropyl alcohol or a suitable alternative. This setup minimizes air exchanges, preserving a contamination-free workspace.

Inoculating Agar Plates: Steps to Follow

  1. Preparation: Ideally, use a flow hood or SAB for this process to mitigate air contamination.
  2. Inoculation Techniques: Depending on your material, such as spore syringes, spore prints, or liquid cultures, various techniques like droplets or scrapings can be employed.
  3. Sealing with Parafilm: When using Parafilm, handle with care to avoid tearing. A systematic approach around the agar plate ensures an airtight seal.

Incubation Insights:Most fungal species thrive at room temperature. However, specific species might benefit from incubation chambers to optimize germination conditions. Be patient; observing growth from spores can span 6-8 weeks. As growth progresses, signs of sectoring might emerge, suggesting genetic variations within the culture.

Detecting Contamination:Distinct characteristics differentiate mycelium from contaminants:

  • Mycelium: Radiates as white, fuzzy growth from the inoculation point.
  • Yeasts/Bacteria: Manifest as white or yellowish milky spots.
  • Molds: While resembling mycelium, they exhibit rapid spread, often turning green or black.

Agar-to-Agar Transfers: A Vital ProcessAs the mycelium encompasses about 80-90% of the agar plate, consider transferring to fresh plates. This approach, especially when targeting uncontaminated sections, facilitates obtaining pristine cultures. The methodical transfer procedure involves careful selection, sterilization, and placement, all within the confines of a sterile environment.

Storage and Maintenance:For prolonged storage, keep agar plates in a sealed container within your refrigerator. This helps in slowing down growth while preserving the integrity of your clean culture.
